Yes — reverse genetics does use reverse transcriptase in certain contexts, because reverse transcriptase is the enzyme that enables the key step of converting RNA into DNA.

In reverse genetics, the goal is to start with a known DNA sequence and work backward to determine the corresponding RNA and protein sequences, or to generate a functional RNA/DNA molecule from a known sequence. One common application is reverse transcription PCR (RT‑PCR), where an RNA sample is first converted into complementary DNA (cDNA) using reverse transcriptase. This cDNA can then be amplified and sequenced to identify the original RNA sequence Thermo Fisher Scientific+1.
